Systems approach to the study of drug transport across membranes using suspension cultures of mammalian cells. I. Theoretical diffusion models.
نویسندگان
چکیده
Some general physical models are described for the diffusional transport of drugs across membranes of cells in culture suspensions. The models provide a basis for the design and analysis of experiments that are aimed to describe (a) the nature of the principal transport barrier, (b) the kinds of drug species being transported, (c) whether, where and how much solute binding occurs, and (d) the influences of pH, partition coefficient and numerous other factors. The cell is treated as a sphere with non-homogeneous phase compartments. Both rigorous and approximate mathematical expressions have been derived for the quasi-steady-state diffusion through the membrane followed by three cases accounting for the distribution of drug jn the heterogeneous cell interior, that is, (a) the non-steady-state situation, (b) establishment of instantaneous distribution and (c) instantaneous distribution in the aqueous interior with slow permeation of drug into the cytoplasmic bodies and nucleus.

A physical model is described for the simultaneous enzymatic bioconversion of a nonelectrolyte solute and the passive transport of both the solute and product of the enzymatic reaction out of cells in culture suspension. The plasma membrane is assumed to be the rate-determining transport barrier. This model provides the basis for the experimental design and analysis of the Michaelis-Menten kine...

The uptake and transport of 9-nitrocamptothecin (9-NC), a potent anticancer agent, across Caco-2 cell monolayers was studied as a free and PLGA nanoparticle loaded drug. Different sizes (110 to 950 nm) of 9-nitrocamptothecin nanoparticles using poly (lactic-glycolic acid) were prepared by via the nanoprecipitation method. The transport of nanoparticles across the Caco-2 cell monolayer as a func...

متن کاملPluronic as nano-carier for drug delivery systems
A common approach for building a drug delivery system is to incorporate the drug within the nanocarrier that results in increased solubility, metabolic stability, and improved circulation time. However, recent developments indicate that selection of polymer nanomaterials can implement more than only inert carrier functions by being biological response modifiers. One representative of such mater...
متن کاملTRANSPORT ROUTE PLANNING MODELS BASED ON FUZZY APPROACH
Transport route planning is one of the most important and frequent activities in supply chain management. The design of information systems for route planning in real contexts faces two relevant challenges: the complexity of the planning and the lack of complete and precise information. The purpose of this paper is to nd methods for the development of transport route planning in uncertainty dec...
